ezMiniPrep is designed for quickly preparing plasmid DNA from small batch of E.coli. You finish the whole process only in 1 tube for each sample and label your tubes once. It works really fast for large batch of miniscreen samples.

  1. Directly pour ~1.35 ml of 37oC, overnight-gown E. coli cell broth in LB or TB to a clear microtube in the kit to lyse cells. (Do not centrifuge)
  2. Invert the tube 3 times and keep the tube for 5 minutes at room temperature. Meanwhile prepare a water-bath to boil.
  3. Place the tube in boiling water for 1.5 minutes and invert the tube at least once.
  4. Immediately centrifuge the tube for 2 minute at 12,000g (~ 13,000 rpm on a typical bench-top centrifuge).
  5. Remove the pellet by using a toothpick provided in the kit.
  6. Add 50 μl of Solution A(blue tube), mix and keep the tube on bench for 3 minutes.
  7. Centrifuge for 3 minutes at 12,000g.
  8. Discard the liquid gently; remove the last drop of liquid on the tube opening by touching the tube opening on a paper tower.
  9. Add 200 μl of Solution B (red tube), tap and keep the tube at room temperature for 1 minute to dissolve the pellet  completely.
  10. Add 400 μl of 95% ethanol and keep the tube at room temperature or -20oC for at least 3  minutes.
  11. Centrifuge for 3 minutes at 12,000g.
  12. Add 200 μl of 70% ethanol to the tube followed by 1 minute of centrifugation at 12, 000g. Then drain the liquid as much as possible. The resulting DNA pellet can be visible or not depending on the copy number of a plasmid.
  13. Dry the DNA pellet. Then dissolve it in 25 μl of TE provided (yellow tube). For restriction, up to 7 μl can be taken for a 10 μl of reaction.

Other notes:
1. Store the kit @ 4C.
2. Leave the kit on bench for few minute before working if any percipitates in Solution B.  
3. Immediately store the unused DNA sample at -20oC after finish preparation.
